The main task of this project is to enhance and strengthen the cross-border research and innovation capacities in the framework of cooperation programme between the two regional research institutions in Upper Austria and South Bohemia that deal with current issues of European and global research. The project task conforms to the regional Innovation Strategy (RIS) of South Bohemia which considers biotechnology as one of the key potentials of the region. The proposed project is also in compliance with other strategic documents of the Czech republic. In Upper Austria it conforms with the Program Innovatives Oberösterreich 2020,  in the section concerning energy: renewable energy /waste streams / biogenic processes, use as value added product or energetic use of biomass. Innovation will be achieved in the field of renewable resources of sustainable energy and valuables compound production from microalgae using biotechnological and genetic engineering. The project is predominantly focused on basic research to enhance scientific knowledge in the production of carbon-neutral sources of raw materials and biofuels that will contribute to higher future independence from fossil sources as well as reduction in atmospheric CO2 levels. 

    

 

Algenetics – project progress 2018-2019

Optimization of growth regime for wild type Synechocystis PCC 6803 was tested in various photobioreactors in the range from 100 mL up to pilot scale of 100 litres. We carried out the possibility of mass cultivation in 20-L flat-plate photobioreactor with fully-automated growth regime for potential production of transformants. Various growth tests were carried out as to induce overproduction of storage compounds (glycogen) by nutrient starvation and increased salinity. The main aim of this project is to construct and test Synechocystis transformants via gen manipulation which overproduce bioethanol to media, or glycogen and starch in biomass as a source of high-energy compounds. The Austrian partners were preparing fully segregated transformants of Synechocystis. Already three series of transformants were prepared: (i) transformants with overexpressed genes glgC to overproduce Glucose pyrophosphorylase, (ii) transformants with overexpressed genes for glycogen synthases glgA1, glgA2 to produced increased amounts of glycogen, (iii) transformants with exchanged Eukaryotic branching enzyme glgB to produce starch-like polysaccharide instead of glycogen, (vi) transformants with overexpressed Pyruvate decarboxylase (pdc) and Alcohol dehydrogenase (adh) genes to overproduce ethanol. These transformants are now tested in laboratory experiments. April 2019
